I was let go from job due to a job mistake involving multiple test sites for hours. I have good references even my boss and boss's boss wanted me to stay but upper management said I had to go. I know if anybody calls HR they won't explain reason but will say we won't rehire this person. How should I be answering questions about why I left that company?

The best way to handle situations like yours is to be honest about why you were terminated. You cannot justify or blame, but will need to show how you have learned from the mistake. Unfortunately, it may not be easy to overcome, eventually there will be a company will be willing to give you an opportunity. When you fill out an application, instead of putting HR as the contact, you should try putting your direct supervisor. This may help your case if the reference is given directly rather than the generic response from HR.
